Abstract
IN AN EARLIER paper, one of us (S. W. S.) reported1 on the use of a combination of an antihistaminic drug and penicillin in an attempt to decrease the allergic responses to the antibiotic. Crystalline potassium penicillin G was dissolved in an isotonic solution of sodium chloride to which had been added 5 mg. per cubic centimeter of doxylamine ( decapryn® ) succinate (2- [α- (2-dimethylaminoethoxy)-α-methylbenzyl] pyridine succinate). Each cubic centimeter of this preparation contained 100,000 units of penicillin and 5 mg. of the antihistaminic drug. In a series of 292 patients, the reaction rate with the use of this doxylamine-penicillin mixture was 2.4 per cent, while in 400 control patients receiving the same aqueous crystalline penicillin alone, the rate was 6.5 per cent.
