Abstract
By autolyzing washed bacterial suspensions (Bacterium coli) in phosphate buffer, Ph 7.6, a dehydrogenase is extracted; by repeated centrifuging and filtration through kieselguhr this may be obtained in clear aqueous solution free from other dehydrogenases of the cell. This preparation, in pres ence of methylene blue, actively dehydrogenates lactate and, much more slowly, anhydroxybutyrate; it is inactive toward all other H donators tested, e.g., glucose, succinate, formate, etc. It differs from the intact cell not only in specificity but in the fact that it can not reduce molecular O except in presence of a carrier such as methylene blue.
