Abstract
In order to study bone marrow reserve, leukopoiesis and leukocytosis, leukocyte desoxyribose nucleic acid (DNA) incorporation of P32 was employed in conjunction with the previously described technique of leukopheresis. A new method of DNA extraction with sodium xylene sulfonate was used. The majority of cells contributing to a leukocytosis induced by leukopheresis are labeled cells released from areas of storage. Evidence is presented that the chief source of the mature leukocytes in such an acute leukocytosis appears to be the marrow reserve. Increased production of leukocytes by acceleration of leukopoiesis occurs only after considerable delay which, in the dog, is about 3-4 days.Keywords
