A Selective Staining Method for the Basic Proteins of Cell Nuclei

Abstract
A staining procedure for a chromosomal protein fraction is described. The method consists in staining formalin-fixed tissue sections after hot trichloracetic extraction with an aqueous solution of the acid dye Fast Green at pH 8. Model experiments indicate that only proteins with relatively high isoelectric points bind dye under the specified conditions. The histological staining picture is presumably due to histone-like proteins. Microspectrophotometric determinations of this protein fraction, in relation to diribonucleic acid measured by the Feulgen reaction, are reported for nuclei of various cell types.Keywords
