Photoactive Subunits of Protochlorophyll(ide) Holochrome

Abstract
A stable, soluble, and photoactive protochlorophyll(ide) complex hasbeenextracted fromdark-grown barley (Hordeum vulgare L.)leaves withbuffer containing saponin andglycerol Afterammoniumsulfate precipitation, theredissolved pig- mentcomplexwaspartially purified bychromatography on Sephadex gelsinthepresence ofsaponin. Withtheassump- tionsthatthepigmentcomplexfrombarley hasthesame shapeanddensity astheproteins usedforcalibration, its molecular weight is63,000. Photoactive protochlorophyll(ide) complexisolated frombean(Phaseolus vulgaris L.)andchro- matographed bythesameprocedures hasanaparent molecu- larweightof100,000orgreater. No chromatographic sepa- rationof photoactive and inactive protochlorophyli(ide) complexes was observed. Photoconversion of protochloro- phyll(ide) tochlorophyli(ide) didnotchangethechromato- graphic behavior ofthepigment complex. Theprotochlorophyll(ide) complexhadanabsorption maxi- mum at644nanometers anda corresponding fluorescence emission maximum at 652 nanometers. Photoconversion yielded chlorophyll(ide) complexwithanabsorption maxi- mum at678nanometers anda mainfluorescence emission maximumat685nanometers. Spectrofluorometry onpartially photoconverted preparations gaveno indication ofenergy transfer fromprotochlorophyll(ide)
