An Improved Method for Culturing Tradescantia Pollen Tubes for Chromosomal Analysis

Abstract
Using pollen of Tradescantia paludosa, growth was greatly enhanced and mitosis accelerated in a medium containing Ca, Mg and K ions, at a high pH. The Ca-supplemented medium, unlike conventional pollen growth media, caused the mitotic chromosomes to align end to end in the pollen tubes. This improved distinctness, and led to more analyzable chromosomes in the pollen tubes.
