Abstract
The establishment of a callus culture of Vicia faba root tissue growing on a modified Bonner and Devirian medium supplemented with 10-6 M 2,4-dichlorophenoxyacetic acid and 5.0 g/l yeast extract is reported. Experiments have been carried out to determine the optimum growth conditions of the callus on semisolid media and an investigation made of the potential of this material as an initial source of friable tissue yielding cell suspensions on dissociation in liquid medium. The development of two morphologically distinct types of callus tissue was observed on semi-solid medium and chemical analysis has demonstrated a correlation between the friability and cell-wall composition of these tissues.Keywords
