Simultaneous Gas Chromatographic Determination of Cocaine, Methadone, Methaqualone, Phencyclidine, and Propoxyphene

Abstract
A gas liquid chromatographic procedure has been developed for the simultaneous identification of methadone, primary metabolite of methadone (2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidine), propoxyphene, norpropoxyphene (primary metabolite of propoxyphene), phencyclidine, methaqualone, and cocaine from small volumes of urine. The drugs and metabolites are extracted from basic urine, converted to acid salts, evaporated to dryness, reconstituted, then injected into a gas chromatograph equipped with a 2% UC W-98/1% OV-17 column. An internal standard (SKF-525-A) is included to monitor the extraction and to allow quantitation of positives. The detection limit of the method for all drugs and metabolites is 0.5 µg/ml.
