Method for Preparation of Complement-fixing Antigens in Study of Experimental Tsutsugamushi Disease (Scrub Typhus)

Abstract
The authors describe a method for preparing complement-fixing antigens of R. orientalis (scrub typhus) from infected yolk sac membranes of fertile hens eggs involving the conc. of anti-genic components by freezing and drying the material prior to removing undesirable lipids by a variety of solvents. The effect of various inactivating agents, lipid solvents and preserving agents is discussed. Advantages of processing antigens in the dried state are: utilization of membranes possessing moderate or even poor rickettsial growth; utilization of lipid solvents miscible or immiscible with water; conc. of antigenic components without resorting to further physical or chemical procedures. The antigen does not exhibit cross- fixation with serums of animals infected with other rickettsial diseases. The method is applicable to large scale production of antigens of R. orientalis and other rickettsiae.Keywords
