INHIBITIONS BY OUABAIN AND PROTOVERATRINE ON ACTIVE TRANSPORT OF L-DOPA INTO BRAIN SLICES

Abstract
Accumulation of dihydroxyphenylalanine L-DOPA in brain slices was inhibited by ouabain, protoveratrine and ethylenediamine tetraacetic acid (EDTA). By addition of cocaine, inhibiting effects of protoveratrine and EDTA on L-DOPA accumulation disappeared but inhibition by ouabain was scarcely influenced. Protoveratrine did not show any influence on the activity of Na+, K+-activated adenosinetriphosphatase (ATP-ase), though ouabain caused a stron inhibition on it. Transport of L-DOPA into brain slices having a low K+ content was slower than the control. From these results, it was speculated that both K+ concentration in brain slices and ouabain sensitive site including Na+, K+-activated ATP-ase might be independently important factors to regulate the active transport of L-DOPA into brain slices.Keywords
