The Effect of the Amount of Nucleic Acid Load on Hydroxylapatite Chromatography

Abstract
The effect of the nucleic acid to hydroxylapatite load ratio (μg/g) has been studied. It was found that as the load ratio was increased from 1 to 100 μg/g there was little change in the elution molarity of the nucleic acids whereas above 1000 μg/g the change could be substantial. Different hydroxylapatite preparations and different nucleic acids gave similar results but the load ratio dependence of the elution molarity was greater at 70 °C than at room temperature.
