Laboratory Suggestion: Two Technics for Permanent Staining of Formalin-fixed Protozoan Cysts

Abstract
Several methods were examined to determine a reliable means of preparing permanent slides of protozoan cysts from formalin-fixed fecal specimens. These included five means of adhering the specimen to the slide and four methods of staining. Two methods of adhering, polyvinyl alcohol infiltration and Schaudinn's fixative with an albumin film, were successful. Two staining methods, iron-hematoxylin and a new acid fuchsin-green stain, were found to give good results after the cysts had adhered to the slides. These technics allow for concentration of formalin-fixed specimens and their subsequent staining for diagnosis, teaching, or reference work.Keywords
