Separation and Estimation of ITa-Estradiol12

Abstract
Evidence for the specificity of Haenni’s iron-Kober reagent B for 17α-estradiol permitting the estimation of this estrogen in the presence of 17β-estradiol has been confirmed and extended. Other naturally occurring estrogens did not react with this reagent. Treatment of urine with β-glucuronidase and dilute acid and purification of the extracts by silica gel chromatography permitted the detection and estimation, as well as providing information regarding the mode of conjugation, of small amounts of 17α-estradiol in urine of the pregnant cow and of the rabbit treated with estrone. While 17α-estradiol was not detected in pooled specimens of human pregnancy urine, evidence was obtained of its presence in the case of one individual.
