On the Concept of Slowly and Rapidly Acting Coumarin Drugs

Abstract
The speed of induction of effect of various anticoagulant drugs was studied in dogs after administration of doses, sufficiently large to block completely synthesis of the relevant clotting factors. No significant difference was found between the latent period for Phenprocoumon, Nicoumalone and ethyl-biscoumacetate. After administration of the 3 different anticoagulants, the clotting factors influenced during coumarin therapy showed the same half-life. Indirect anticoagulants differ in the time during which they are linked to the sites of synthesis of the involved clotting factors. Their rapidity of action on clotting factors is dependent on dosage and not on chemical properties.Keywords
