Theory of Preparative Linear Chromatography by Elution Development: The Basic Equations

Abstract
An equation has been derived to describe the efficiency of a preparative-scale chromatographic column in terms of the factors which influence it. The effect of the volume of the sample at the column inlet on the amount of material of a given purity which may be recovered per unit time has been investigated. It has been shown that, under all circumstances, there is an optimum value of the sample inlet volume.
