Relationship between neurological symptoms and concentrations of clioquinol in serum and nervous systems of beagle dogs.

Abstract
A fixed dose of clioquinol, 300 mg/kg per day, was orally administered in gelatin capsules to beagle dogs. Powdered clioquinol, 300 mg/kg per day after increasing dose, was orally administered in milk twice a day to another group of dogs. All dogs administered clioquinol developed hind-limb weakness and hip swaying in 10 days. Individual differences and daily variations in serum clioquinol, its surface and its glucuronide levels were large. Clioquinol was detected at higher concentrations not only in CNS but also in peripheral nervous system and was retained for a long time.
