A Method for the Determination of Tracer Phosphate in Biological Material.

Abstract
The method of Martin and Doty (Analyt. Chem. 21: 965. 1949) for orthophosphate is modified whereby the derivatives of phosphomolybdic acid formed are extracted with a mixture of butanol and benzene (1:1). The color of the extract is detd. at 640 m[mu]. The specific activity of the phosphate is obtained by direct radioactivity measurement of an aliquot of the extract. The original sample is usually 3 ml. containing 2-16 [mu]g. P. The amt. and specific activity of creatine phosphate may also be detd. on the same sample. The above method overcomes erroneously high orthophosphate values obtained by precipitation methods in the presence of mixed phosphates.Keywords
