Measurement of Osmotic Fragility of Fibroblasts

Abstract
Resistance of cells to disruption due to swelling in hypotonic solutions was used as a measure of cell membrane strength. Earle''s strain L mouse fibroblasts disrupted at 13% of isotonicity compared with 50% of isotonicity for erythrocytes. Pretreatment of fibroblasts with trypsin or papain did not decrease their resistance to osmotic lysis, suggesting that the mechanical strength of the fibroblast cell membrane does not depend on a protein substrate of these enzymes.
