A STUDY OF THE ANTICHOLENESTERASE AND ANTICURARE EFFECTS OF SOME CHOLINESTERASE INHIBITORS

Abstract
Inhibition of serum cholinesterase by neostigmine, tacrine, pyrido- stigmine and edrophonium was measured in vivo under clinical conditions. In vitro inhibition of serum and red blood cell chollnesterases by these agents was also studied. Anticurare action of neostigmine and tacrine was compared myographically. Previous clinical observations are confirmed and support is given to the theory that neostigmine has a direct action on the neuromuscular junction or muscle fiber, apart from its anticholinesterase property. However, the degree of enzyme inhibition depends on concentrations of enzyme, substrate and inhibitor; since levels of these at the neuromuscular junction are unknown, they are not reproducible for experimental study.
