On the Mechanism of Action of Tolbutamide

Abstract
The influence of intravenous tolbutamide, given alone or following a single injection of insulin or tolbutamide, has been studied in normal dogs. Parameters measured were blood glucose, plasma free fatty acids and plasma immunoreactive insulin. A reciprocal relationship was seen between hypoglycemia and rebound lipolysis with either a single insulin or a single tolbutamide injection whereas the repetitive administration of tolbutamide resulted in suppression of lipolysis despite sustained hypoglycemia. These results allow the suggestion that the chronic effect of tolbutamide may be related to enhancement of insulin action upon antilipolysis. 1 This work was supported by grant #U-1960 from the Health Research Council of the City of New York and a Research Grant from the American Diabetes Association, New York, N.
