Abstract
Two species of salmon (Oncorhynchus kisutch, O. nerka) and steelhead and rainbow trout (Salmo gairdneri) were injected with Evans blue dye. Typical excretion slopes and a minimum mixing time of 60 min were determined by sampling blood through a dorsal aorta cannula for up to 5 hr after injection. Extrapolation of the excretion slope to zero time provided an estimate of the dye dilution which, with hematocrit measurements, gave blood volume estimates ranging from 5.4 ± 0.8 to 6.9 ± 1.8 ml/100 g.A new method of using Evans blue was tested in which dye was injected into one fish and its plasma reinjected into other fish. Blood volume estimates for O. kisutch were 4.5 ± 1.5 by this method as compared with 6.2 ± 2.0 ml/100 g for Evans blue used in the usual manner in the same individuals.Blood volumes estimated from plasma samples taken 20, 25, and 30 min after injection while the fish were maintained out of water were 35% less than after the same fish were returned to water and a second estimate made from additional samples taken 1, 2, and 3 hr after the original dye injection. Keywords
