A Method for Esterification of Valproic Acid for Gas-Liquid Chromatography: Clinical Data from Epileptic Patients

Abstract
A flame ionization gas-liquid chromatographic (GLC) method is presented for the determination of serum levels of valproic acid (n-dipropylacetic acid, 2-propylpentanoic acid) using cyclohexylacetic acid (cyclohexaneacetic acid) as an internal standard. Methyl ester derivatives of both compounds are formed using 1.25 mol/liter HCl in methanol. The derivatives are separated using a 6-ft. glass column packed with 5% OV-17 on Gas Chrom Q converted to an acid phase. Data from 258 epileptic patient samples is presented. Serum levels of 47% of this group were above 40µg/ml.Keywords
