SEPARATION OF ENCEPHALOMYOCARDITIS VIRUS FROM TISSUE COMPONENTS BY MEANS OF PROTAMINE PRECIPITATION AND ENZYMIC DIGESTION

Abstract
Suspensions of mouse brain infected with encephalomyocarditis virus were treated with protamine sulfate and centrifuged. The supernatant fluid was centrifuged at high speed and the resulting sediment treated with crystalline trypsin. The digested suspension was again concentrated in the ultracentrifuge and the final sediment tested for infectivity and N content. It was possible to obtain suspensions with as much as 1015.9 LD50 per gram of N. The same material contained particles, which are considered to be the virus, characterized by a sedimentation constant of 151 S and a diam. of 27 to 30 m[mu]. A "normal component" with a diam. of about 18 m[mu] was also found in both infected and normal prepns.Keywords
