Glycoconjugate Cytochemistry of the Rat Small Intestine Using Helix pomatia Agglutinin and Colloidal Gold Conjugates

Abstract
The glycoconjugate localization of the rat jejunal and ileal tissues was examined by the postembedding staining method using Helix pomatia agglutinin (HPA) and colloidal gold (CG) conjugate and ultrathln sections obtained from a hydrophilic resin Lowicryl K4M. HPA-CG conjugates were observed over the microvillous region of both columnar epithelial and goblet cells, carrier vesicles of the columnar epithelial cells, the Golgi apparatus of these two types of cells and in the granules of the Paneth cells. This method is easy, not time-consuming and can be used for localizing sugar residues of the cell surface and cytoplasm in relation to cell organeUae, especially the Golgi apparatus.
