Effect of hydrogen ion concentration and of temperature on the growth ofParacoccidioides brasiliensisin soil extracts

Abstract
The survival of Paracoccidioides brasiliensis in soil extracts adjusted to various pH values and subjected to different temperatures was investigated. The mycelial phase was more resistant to extreme conditions than the yeast phase. Both phases were inhibited by pH 1·4; the yeast phase did not survive pH 3·0 but many of the mycelial elements were viable. With both phases, an increase in the number of viable particles occurred in the extracts with initial pH values of 7·0 and 5·0. However, growth of the mycelial phase was maintained for longer periods. Both types of elements altered the original pH values of the soil extracts in order to reach slightly acidic values (6·2-6·9). Neither phase survived exposure for 3 weeks at — 20° C. or at 45° C. The mycelial elements tolerated 4° C. and were even able of growth at 15° C. while the yeast cells were more sensitive to the effect of the various temperatures. These results are discussed in relation to the chances that each type of cell would have for survival saprophytically in nature. Se estudió la capacidad de supervivencia del Paracoccidioides brasiliensis en extractos de suelos con variados pH y sometidos a la acción de diferentes temperaturas. La fase micelial demostró ser más resistente a las condiciones extremas que la levadura. Ambas fases se vieron inhibidas por pH 1·4; las levaduras no sobrevivieron tampoco pH 3·0 pero un porcentaje apreciable de las formas miceliares eran aún viables al finalizar el experimento. Los extractos de suelo con pH 5·0 y 7·0 permitieron un aumento del número de células viables tanto de la fase levadura como de la fase miceliar. Sin embargo, el crecimiento de esta última fué más prolongado. Ambas fases ocasionaron cambios en el pH original de los extractos, acercándolos a los valores aparentemente óptimos. (pH 6·2–6·9). Con respecto a la temperature, ninguna de las fases sobrevivió la exposición por 3 semanas a -20° C. ó a 45° C. Los elementos miceliares toleraron 4° C. y además, fueron capaces de crecer al 15° C. Las levaduras, en cambio, se mostraron mucho más sensibles a la acción prolongada de las varias temperaturas empleadas. Se discuten estos resultados en relación con las posibilidades de supervivencia que tendría cada fase como forma saprofíta en la naturaleza.Keywords
