Abstract
The paper illustrates the application of some recent technical improvements in thin sectioning to unusually favourable material. By means of lead-stained sections of osmic-fixed material embedded in epon, much new information regarding the fine structure of the flagellar apparatus, including in particular the flagellar ‘roots’ and flagellar bases, has been provided. The sinking in of the flagellar apparatus after secretion of a cell wall has been traced in outline, together with the persistence of parts of the flagellar bases into the two-celled germling in a manner suggesting a centrosomal function. Other observations include details of the structure and arrangement of endoplasmic reticulum in swimming cells as seen in serial sections after both osmic and permanganate fixation, some preliminary observations on rearrangement in position of this component in the early stages of germination, observations on the contractile vacuoles and on the special type of ‘hairy’ vesicle associated with them, on the Golgi bodies in both swimming and settled cells, on the distribution of ribosomes in relation to plastids, flagellar bases, the nuclear envelope, &c., and on the contents, distribution, and possible nature of various types of vesicles, including the pigment chambers of the eye-spot, vesicular derivatives of Golgi bodies, and vesicles beneath the plasmalemma in swimming cells from which the first components of the cell wall are believed to be liberated.
