Amoxycillin: Pharmakokinetik und Erfahrungen bei der Behandlung von Harnwegsinfektionen

Abstract
We investigated the pharmacokinetics and clinical efficacy of amoxycillin which, whilst possessing a similar bacteriological range of activity and similar toxicity, is better absorbed than ampicillin. In 10 young healthy persons, the maximum serum concentration of amoxycillin is 1.8 times higher than the average for ampicillin. The urinary recovery of amoxycillin is also proportionate to the improved absorption, being at 51.9% almost the double of that of ampicillin at 28.3%. In order to test the clinical efficacy of amoxycillin we treated 58 patients with urinary infections, chronic pyelonephritis and superinfections of other kidney diseases. After a 17-day therapy of 3 × 1 g daily, the previously infected bladder puncture specimen was rendered sterile in 38 (66%) out of 58 patients. Furthermore, we observed a significant decrease of leucocyturia from 190.4 to 57.8 millions/24 h. Albuminuria, erythrocyturia and ESR did not appear suited for the continuous assessment of an anti-infective therapy for kidney and urinary diseases. As side-effects we noted slight exanthemata, enanthemata and diarrhoea. Only in one case it was necessary to discontinue the treatment.
