COMPARISON OF ALDEHYDE FUCHSIN STAINING, METACHROMASIA AND PERIODIC ACID-SCHIFF REACTIVITY OF VARIOUS TISSUES

Abstract
A survey of a wide variety of tissues was carried out, comparing their staining with aldehyde fuchsin, the periodic acid-Schiff (PAS) method (after salivary digestion) and toluidin blue metachromasia. The following observations were made: (1) Most of the chromo-tropic substances are also aldehyde fuchsin positive, but fixation abolishing metachromasy does not necessarily diminish the affinity for aldehyde fuchsin. (2) Many orthochromatic, PAS reactive structures stain with aldehyde fuchsin while others fail to do so. At least some of the latter can be made aldehyde fuchsin positive by oxidizing the section in an acid soln. of permanganate prior to staining. This pretreatment also greatly enhances the staining of the neurohypophysial neurosecretory substance. (3) A small and heterogeneous group of tissue elements is aldehyde fuchsin positive, but both orthochromatic and PAS negative. No suggestions can be made regarding the histochemical basis of aldehyde fuchsin staining.Keywords
