Disposition studies with amikacin after rapid intravenous and intramuscular administration to human volunteers

Abstract
Serum and urine levels after rapid intravenous and intramuscular injection of the new aminoglycoside antibiotic amikacin at two dose levels have been measured in 6 young volunteers. Serum levels after a 125 mg dose remained above 4 μg/ml for 2 and 2.5 h for the i.v. and i.m. routes respectively but only exceeded 10 μg/ml for about 30 min after the i.v. injection. Serum levels after a 500 mg dose remained above 10 μg/ml for 3 and 5 h after the i.v. and i.m. injections respectively. Pharmacokinetic analysis shows that serum amikacin clearance is less than the glomerular filtration rate, that the AUC is proportionately greater after i.m. administration and that the apparent serum half life is between 2.5 and 5 h. Seventyfive to eighty per cent of the administered dose was recovered within 12 h in urine and the rate of urinary excretion was proportional to the serum amikacin concentrations.Keywords
