Abstract
A double antibody radioimmunoassay is described which is capable of detecting 100 pg of medroxyprogesterone acetate (Provera) in 0.1 ml of unextracted serum. The antibody was produced to steroid conjugated at the 3 position with bovine serum albumin. The side groups attached at 17 and 21 positions are more important determinants of antibody specificity than groups attached at 6. No significant crossreactivity was noted with physiological concentrations of naturally occurring steroids. The concentration of Provera in peripheral serum was determined for female monkeys and humans that received the drug by various routes of administration. A rapid decrease to less than one half the initial concentration was noted within 10 min after an intravenous injection. Blood levels were maximum at 2 or 4 hr after oral administration. Intramuscular injection resulted in fluctuating blood levels that were maximal at 2–15 days post injection. Following insertion of a Provera silastic vaginal device, consistently low blood levels were found during the 21 days that the device was in place.Keywords
