Abstract
NG108‐15 cells were transfected to stably express a haemagglutinin epitope‐tagged variant of the long isoform of Gsα. Clone BST15 expressed this polypeptide at similar levels to the endogenous long isoform of Gsα. Treatment of clone BST15 with the IP prostanoid receptor agonist iloprost resulted in down‐regulation of both forms of Gsα with both dose—effect curves to iloprost and time courses of loss of the two forms of Gsα being indistinguishable. These results demonstrate that the IP prostanoid receptor interacts with and regulates the epitope‐tagged variant of Gsα in an equivalent manner to the unmodified protein and indicates that the epitope‐tagged polypeptide can be used to analyse mechanisms of receptor regulation of cellular G‐protein levels.
