Gene Transfer System for Potato

Abstract
A foreign gene encoding neomycin phosphotransferase (Nos/Npt), specifying resistance to kanamycin, was introduced into potato cells via a disarmed Agrobacterium tumefaciens. The Agrobacterium was incubated with leaf-derived callus followed by selection with 50 mg°liter−1 kanamycin. Neomycin phosphotransferase (NPT) conferred on the transformed potato cells the capacity to grow in the presence of kanamycin. Enzymatic assay demonstrated that the kanamycin-resistant calli contained NPT activity whereas the sensitive calli were devoid of this activity. Transgenic plants were recovered from these resistant calli and the foreign gene was expressed in all tested tissues of these plants as well as in tubers and developing sprouts.Keywords
