Abstract
In the prepn. of T3 Escherichia coli bacteriophage it was found that liter-scale batches failed to reproduce the results of small scale (10 ml.) expts. because of differences in aeration efficiency. An all-glass apparatus is described which is based on an air-lift circulator with a cyclone-type gas dis-engager. It has no moving parts but permits large air flows with excellent agitation. External resistance windings allow sterilization in situ, and provide temp. control. Cotton inlet and effluent filters largely avoid the problems of air-borne contamination which had been previously experienced. With an apparatus of this design the results of small-scale expts. have been reproduced on a 2- and 6-liter scale.Keywords
