Abstract
These studies indicate that echothiophate iodide is capable of producing marked inhibition of brain cholinesterase on chronic administration as eye drops, in rabbits. Demecarium bromide produced relatively little inhibition of brain cholinesterase. Isofluorophate produced little systemic inhibition of enzyme activity, probably because of hydrolysis or defluoridation of the compound. Marked inhibition of enzyme activity was found with demecarium, echothiophate, and Paraoxon, and most of this effect occurred within the first 7 days of treatment, so that little increment of inhibition occurred in subsequent treatment up to 21 days. No symptoms of muscarinic or nicotinic toxicity were noted, but 2 rabbits died during the experiment while under treatment with echothiophate.
