Abstract
A technique is described for the large-scale culture of sycamore cell suspensions. Two 10-1 wide-mouthed flat-bottomed culture bottles are spun continuously at 120 rpm inclined at 45° from the vertical. Each bottle contains a 4.5 1 culture which, after 21 d at 25 °C yields a suspension of 109 cells, 1.4 1 of packed-cell volume and 40 g dry weight. The growth characteristics of the cultures are described and it is demonstrated that large volumes of suspension can be withdrawn during incubation without altering the general growth pattern or the yield of cells per unit volume of the final culture.Keywords
