USE OF GLASS BEADS FOR THE MECHANICAL RUPTURE OF MICROORGANISMS IN CONCENTRATED SUSPENSIONS

Abstract
Pavement marking beads (sieve size 60/80) can be used in the Waring blendor and in tissue homogenizers to rupture slurries or con-centrated suspensions of bacteria and yeasts. The efficiency of disruption is controlled by the relative amts. of beads and suspension. With a fixed total number of organisms, the smaller the vol. of suspending liquid, the greater the rate of cell destruction. Likewise, for a given vol. of suspension, efficiency is improved by increasing the quantity of beads, subject only to the limitation that the stirring device be capable of vigorous agitation of the mixture. Ideally, grinding would appear to be most extensive when the volume of suspension employed is just sufficient to occupy the spaces between the beads present.
