STERILIZATION TECHNIQUE FOR GRASS SEEDS

Abstract
The method which this author has used for sterilizing many kinds of plant seeds in obtaining sterile seedlings for aseptic culture includes treatment with Semesan, an organic Hg compound, the active fraction of which is hydroxymercurichlorophenol. The effects of this substance, however, were lethal to grass seeds, as were those of all other substances tested containing Hg. Of the additional substances tested only Ca hypochlorite appeared to produce no physiol. injury, but it was found necessary in order to obtain adequate sterilization to first wet the seeds with 95% alcohol. Soaking the seeds for 30 min. in a 10% (filtered) Ca hypochlorite soln. appeared to be the most desirable procedure.
