Abstract
Probenecid, when administered s.c. to F344 rats 1 h prior to an i.v. LD50 dose of cis-dichlorodiammineplatinum(II) (CDDP), reduced significantly the peak BUN [blood urea nitrogen] and serum creatinine levels which occurred 4 days following CDDP. Renal histopathology was similarly reduced by probenecid pretreatment was 17 mg/kg; the LD50 value of CDDP administered i.n. without probenecid pretreatment was 17 mg/kg; the LD50 dose with probenecid was increased to > 19 mg/kg. Probenecid pretreatment did not influence the therapeutic response to CDDP when the latter was administered to BDF1 mice bearing L1210 Leukemia.
