Quantification of Serum IgM in Pigs by Monoclonal Antibody: Enzyme‐Linked Immunosorbent Assay, “Dot Immunobinding” Assay, and Radial Immunodiffusion

Abstract
Summary: Monoclonal anti‐IgM antibody (LIG 4) was used in enzymoimmunological test (ELISA) for accurate determination of IgM level in pig sera from birth up to the age of two months and in the sera of various pig breeds and crossbreds. Optimum test conditions as well as errors of the method were determined. For a rapid, sensitive and simple estimation of porcine IgM in serum, a modified semiquantitative dot immunobinding assay on nitrocellulose sheets is recommended. A good correlation between this universal technique and ELISA was established. In addition the amount of IgM was determined using radial immunodiffusion (RID) and the LIG 4 precipitating antibody. The minimal amount of IgM detected by the ELISA was 1 ng, by the dot immunobinding assay 2 ng and by radial immunodiffusion 300 ng in 100, 2 and 10 μl of pig sera, respectively.Zusammenfassung: Die Quantifikation des IgM im Schweineserum mittels monoklonaler Antikörper: Immunoenzymatischer Test (ELISA), “Dot Immunobinding”‐Test und radiale ImmunodiffusionMonoklonale Antikörper gegen IgM (LIG 4) wurden in einem ELISA zur genauen Bestimmung der IgM‐Konzentration in den Seren von Schweinen verschiedener Rassen und Kreuzungen von der Geburt bis zum Alter von 2 Monaten verwendet. Optimale Testbedingungen sowie Fehlerquellen wurden herausgefunden. Zur schnellen, empfindlichen und einfachen Bestimmung der Konzentration des IgM im Schweineserum wurde ein halbquantitativer “dot immunobinding”‐Test auf Nitrozelluloseblättchen modifiziert. Eine gute Korrelation zwischen dieser universal verwendeten Technik und dem ELISA wurde festgestellt. Zusätzlich wurde die IgM‐Konzentration mittels radialer Immunodiffusion (RID) mit dem LIG 4 als präzipitierenden Antikörper bestimmt. Die Mindestkonzentration von IgM, die mit dem ELISA bestimmt werden konnte, war 1 ng in 100 μl Schweineserum, mit dem “dot immunobinding”‐Test 2 ng in 2 μl und mit radialer Immunodiffusion 300 ng in 10 μl Schweineserum.Keywords
