Abstract
An alginate lyase from the culture medium of Alteromonas sp. H-4 was purified by ultrafiltra-tion, gel filtration, and anion-exchange chromatography and was characterized. A molecular weight of the purified enzyme was estimated as 32, 000 by sodium dodecyl sulfate-polyacrylamide gelectrophoresis (SDS-PAGE). Optimum pH and temperature of the enzyme activity were 7.5 and 30°C, respectively. The enzyme was unstable on heating and in acidic solution. The al-ginate lyase required 50mm MgCl2 or MgSO4, 0.5M NaCI, and 0.2M KCl for the maximum acti-vityKeywords
