Carbon Isotope Discrimination of Epidermal Tissue and Mesophyll Tissue from the Leaves of Various Plants

Abstract
δ13C values (carbon isotope discrimination ratios) of epidermal and mesophyll tissues from leaves of a number of plant species were measured. Values for the epidermal tissue were marginally, but consistently, less negative than values for the mesophyll tissue (difference significant at P < 0.002 level). All values, however, were typical of C3 plants. This suggests that metabolites from the mesophyll are transported to the epidermal layer and that PEP carboxylase does not make a large contribution to the metabolism of epidermal tissue. δ13C values for the ethanol/water-insoluble fraction of tulip leaf mesophyll and epidermal tissue were less negative than the ethanol/water-soluble fraction; this was particularly evident for the epidermal tissue (−22.7% and −25.4% for the insoluble and soluble fractions, respectively). The significance of this has yet to be determined.Keywords
