Porcine pancreatic prokallikrein. IV N-terminal amino acid sequences of prokallikrein B and the kallikrein generated from it by the action of trypsin.

Abstract
Porcine pancreatic prokallikreins A and B were separately highly purified by water extraction in the presence of various protease inhibitors, ammonium sulfate salting-out fractionation, anion-exchange chromatography, immunoaffinity chromatography on an anti-porcine pancreatic kallikrein B antibody immobilized Sepharose 4B column and gel filtration on Sephacryl S-200. The specific activities of the final prokallikreins A and B preparations were 75.0 and 79.4 EU/A280 (when assayed after activation with trypsin), respectively, and the prokallikrein B preparation showed a single band on the sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The N-terminal amino acid sequence of prokallikrein B was H-Ala-Pro-Pro- and that of kallikrein B", which is the kallikrein generated from prokallikrein B by the action of trypsin, was H-Ile-Ile-Gly-Gly-. The latter sequence agreed with that of the A-chain of kallikrein B, which is the kallikrein obtained from the autolyzed pancreas.
