The Formation of Germ Tubes byCandida albicansin Sheep Serum and Trypticase Soya Broth

Abstract
Twenty-five strains of Candida albicans isolated from human material all developed germ tubes when incubated for 3 hours in sheep serum diluted up to 1:32 either with saline solution, or with peptone water, or with trypticase soya broth. Seventy-nine strains of Candida albicans all developed germ tubes after incubation for 2 hours in 100% sheep serum and also after incubation for 3 hours in trypticase soya broth alone, whereas 21 strains of Candida from five species other than Candida albicans did not. It is concluded that either sheep serum diluted as much as 1:32 or trypticase soya broth is a reliable medium for the germ tube test for Candida albicans. However, care should be taken to ensure that the inoculum amounts to less than 106 organisms per milliliter of medium to avoid false-negative tests with strains of Candida albicans with only a slight tendency to form germ tubes.
