Abstract
This is a useful modification of a method previously described by the same Author (Experientia 15:274, 1959). Small pieces of fresh tissue are fixed in 10% formalin or in 1% osmic acid and embedded in paraffin. Sufficiently thin sections (1-4 [mu]) are stained for 5 min in the following solution: Acid fuchsin 0.2 gm; Methyl blue 0.1 gm; 1N Hydrochloric acid 100 ml. Then the sections are rinsed gently in distilled water, deydrated quickly in 95% and absolute alcohol, cleared in xylene and mounted in balsam. Mitochondria stain purple red with peripeheral blue wall, chromatin and collagen blue, nucleoli and erithrocites brillant red.
