Abstract
Melted and inoculated medium is placed in a culture tube of 25 X 200 mm. An electric motor spins the tube vertically in a water bath. The tube is driven by a rubber stopper on the shaft of the motor, fitting the mouth of the tube. The base is seated in a ball step bearing. The medium climbs the tube and the film is solidified instantaneously in this position, by filling the surrounding bath with ice water. Either agar or gelatin is suitable.
