LOCALIZATION OF ACID PHOSPHATASE IN THE ADRENAL MEDULLARY CELL

Abstract
About 70% of the acid phosphatase activity of bovine adrenal medulla is localized in the granular fraction containing the sympathomimetic amines. Unlike the mitochondrial acid phosphatase which was readily liberated from the mitochondria upon lysis, the phosphatase in the medullary granules was bound to some water insoluble structure of the granules since only a portion was released into the media upon lysis in hypotonic solutions. In addition the medullary granules contained an ATP dephosphorylating system which possessed a high activity up to pH 8 and was highly insensitive to NaF. Due to the activity of this system the large amounts of intra-granular ATP were rapidly broken down to AMP on transferring the granules to a hypotonic medium.Keywords
