Handling of Oat Chromosomes1

Abstract
A procedure and schedule for germinating oat seeds and preparing root tip chromosomes is presented. Control of environmental factors that affect germination and growth produced root tips with many metaphase cells. The main features of the procedure are: 1) use of plastic petri dishes and filter paper, 2) 2.65 ml distilled water per petri dish with seven seeds, 3) cold treatment (0‐4 C) of seeds in petri dish for 48 hours, 4) incubation of seeds in 27 C incubator for 44 hours, and 5) pretreatment of root tips in 0.002 M 8‐hydroxyquinoIine at 12‐13 C for 3.67 to 4 hours. Root tips are fixed in Farmer's fluid and kept in a 37 C oven for 48 hours before hydrolysis and staining with Feulgen.This publication has 0 references indexed in Scilit:
