Identification of Sterols on Filter Paper and Their Separation by Paper Partition Chromatography.

Abstract
Cholesterol, ergosterol, 7-dehydrocholesterol, and vits. D2 and D3 were identified on filter paper by treating with Br2 and spraying with KI and starch solns. or by spraying with SbCl5 soln. The solvent combination phenol 13.5 parts by vol., methanol 30, water 56.5, separated 7-dehydrocholesterol from cholesterol and ergosterol on an ascending chromatogram, using 10 ug. quantities of each. With phenol 14, methanol 30 and water 56 parts, 7-dehydrocholesterol and cholesterol migrated together, leaving ergosterol behind. With methanol 80, benzene 10 and petroleum ether 10, 10 ug. each of vit. D3, cholesterol and 7-dehydrocholesterol were separated and distributed in 3 bands from the solvent front on backwards, on a circular chromatogram. Cholesterol could be separated from ergosterol using the same solvent and conditions.Keywords
