Glutathione transferase Omega class polymorphisms in Parkinson disease

Abstract
Within 1 the glutathione transferase (GST) superfamily, the Omega class contains two expressed genes (hGSTO1 and hGSTO2) and one pseudogene (hGSTO3p). 1 The hGSTO1 and hGSTO2 genes lie 7.5 kb apart on chromosome 10q24.3, 1 between the markers D10S603 and D10S597. This region overlaps the D10S1239-D10S1237 interval implicated in age at onset (AAO) of Alzheimer disease (AD) and Parkinson disease (PD). 2 We previously identified three polymorphisms in hGSTO genes: hGSTO1*A140D, hGSTO1*E155del, and hGSTO2*N142D. 1 Omega class GSTs regenerate ascorbate, 3 an important antioxidant in the brain. Additionally, GSTO1-1 may activate interleukin-1[beta] 4 and contribute to inflammation, which is implicated in AD and PD pathogenesis.
