A Simple Plasminogen Determination by a Passive Hemagglutination–Inhibition Technic

Abstract
Plasminogen was direcdy coupled to human O cells by mixing 0.1 ml. of packed cells with 800 μg. of plasminogen, 1 ml. of citrate buffer, pH .6.4, and 0.25 ml. of 2.5% glutaraldehyde in H2O. Coupling was accomplished after rotating at 180 r.p.m. for 2 hours at room temperature. After further washing and titration with antiplasminogen, the preparation is highly suitable for passive hemagglutination–inhibition testing for plasminogen. Serum instead of plasma or the euglobulin fraction can be used. The values obtained compared favorably widi the similar method. Reproducibility was good, and sensitivity of 0.016 CU per ml. was easily obtained. Bypassing the tanning process gready simplifies the procedure.Keywords
